Scooby-Doo and Daphne Blake are very close friends in their mystery-solving group.

Daphne is the first person Scooby tries to replace Shaggy with as a best friend. While both of them are "best friends" they develop a bond due to moments like sharing food and being alone together. Daphne was concerned about Scooby being objectified in a dog show, so agreed to be his coach/handler.

When Scooby-Doo and Shaggy Rogers had a falling out, the former moved onto the other members of the gang, starting with Daphne. Things went well until his bag of chips got soaked, and she didn't share his grief over it.[1]
Daphne became interest in the crystals of the Crystal Canopy being able to make telepathic thoughts, which she then would be able to know what dogs were thinking, to which he responded by saying she wouldn't understand.[2]
When the gang heard about a mystery at a dog show, they have Scooby participate in it to help solve a mystery. Daphne became Scooby's coach for the dog show. Scooby and Daphne went together to search for clues and they performed a well choreographed dance in the dog show. The Houndbeast was about to approach Daphne and hurt her, but Scooby overcame his fears in order to protect her. The other dogs joined in and they all barked and chased it around the arena with the Houndbeast crashing into the judge's table. Scooby won the dog show and put his medal on Daphne's neck and she gave him a loving hug with the rest of the gang. [3]
